Ipamorelin is a five-residue selective agonist at the ghrelin receptor GHS-R1a, characterised in 1998 as the first secretagogue in its class to release growth hormone without a matching rise in cortisol, prolactin or ACTH. 12

Human work on GHRH given together with a growth-hormone-releasing peptide reports a response larger than the sum of the two given separately, and that synergy is the published basis for combining the classes rather than stacking two compounds from one class. 3

The synergy studies used GHRH alongside GHRP-class peptides, not CJC-1295 alongside ipamorelin. No published study has tested this combination, and the 5 mg to 5 mg ratio is a market convention rather than a researched proportion. Whether the same synergy holds for these two particular molecules at this particular ratio is untested.
