BDNF expression
The most-cited Semax finding is increased expression of brain-derived neurotrophic factor in rat hippocampus, where a single application raised BDNF protein and trkB phosphorylation alongside a measurable change in conditioned avoidance behaviour. BDNF supports the survival and growth of neurons, and this is the proposed basis for the cognitive research. 2 A separate line of work reports that Semax raises striatal serotonin turnover and amplifies amphetamine-evoked dopamine release while not altering dopamine on its own, which is a second and largely independent mechanism. 1
Neuroprotection models
Rodent stroke and cerebral ischaemia models are the main setting for Semax research, examining whether tissue damage is reduced when the peptide is administered after the event. 3
No hormonal activity
Although derived from ACTH, Semax covers residues 4 to 10, outside the region that stimulates the adrenal gland. It does not raise cortisol, which is precisely what allows it to be studied as a neuropeptide rather than a hormone analogue.
