Anxiolytic research
Selank was developed from tuftsin as an anxiolytic candidate, and interest persists because it is reported to act without the sedation, tolerance or dependence associated with benzodiazepines. The main published human study compared it directly against the benzodiazepine medazepam in generalized anxiety disorder and neurasthenia, reporting a comparable anxiolytic effect with additional antiasthenic and psychostimulant activity. 1 That is a 62-patient Russian-language trial which has not been independently replicated, and it carries correspondingly less weight than the journal listing alone suggests.
GABA and serotonin systems
Animal work reports changes in the expression of GABA-A receptor subunits and in serotonin turnover, and these are the pathways most commonly proposed to explain the behavioural observations. Rodent work also reports that Selank attenuates the aversive signs of naloxone-precipitated morphine withdrawal, performing close to but below diazepam in the same model. 2
Peptide stability design
Tuftsin itself is cleared almost immediately. Adding Pro-Gly-Pro to the C-terminus slows that enough to make the molecule usable, the same approach taken with Semax. It is a clean illustration of how terminal extensions protect very short peptides.
