GHRH receptor pharmacology

Sermorelin is the 1-29 N-terminal fragment of growth-hormone-releasing hormone, amidated at the C-terminus. Deletion work on the parent hormone mapped where the activity actually sits: shortening hGRF to the 1-29 fragment leaves roughly half the potency of the full-length hormone, fragments as short as 1-21 remain active though at very low potency, and bioactivity only disappears once the chain is cut back to 1-19, with the minimal fully intrinsic core falling in the 3-21 region. 1 That makes 1-29 the shortest fragment still practical as a full agonist rather than an arbitrary cut, which is why it is the length carried forward into the rest of the analogue family.

Short half-life as a research property

Sermorelin retains the Ala2-Asp3 bond of the parent hormone and is cleaved there by dipeptidyl peptidase-4 at essentially the same rate as full-length GRF, which is what gives the unmodified peptide a plasma half-life measured in minutes. 2 That was a practical limitation in its clinical era and is a useful property in a laboratory setting, where a short-acting agonist separates an acute receptor response from the effects of sustained exposure. It is also the specific bond that the later analogues in this family were designed around.

Comparative GHRH work

It is routinely run against longer-acting analogues in the same family: tesamorelin, which is full-length and modified, and the CJC-1295 forms, which are the same 1-29 fragment with substitutions and in one case an albumin-binding group. Because all of them act at the same receptor, the comparison isolates what changing exposure over time does rather than what the peptide does at the receptor. Direct comparisons of the 1-29 fragment against the full-length hormone report differences in how repeatable the response is on re-exposure, not only in potency. 3

What is sermorelin peptide?

Sermorelin is the first 29 amino acids of growth-hormone-releasing hormone, synthesised as a standalone peptide. Truncating GHRH to this length keeps its activity at the receptor; shortening it further does not.
