The collagen fragment hypothesis
KTTKS is released when type I collagen is broken down. The reasoning behind using it as an active is that a fragment of degraded collagen may act as a feedback signal that repair is needed. In tendon-cell culture the pentapeptide increased expression of type I collagen and transforming growth factor beta, which is the most direct published support for the idea. 1

Why is it palmitoylated?
Because KTTKS on its own is hydrophilic and crosses lipid membranes poorly. Attaching a fatty acid makes the molecule amphiphilic and improves partitioning into the stratum corneum in permeation studies.
