Handling

How should PT-141 be stored and reconstituted?

Lyophilised storage. Sealed vials are stable at room temperature for short periods during transit. For longer-term storage keep at −20 °C, protected from light and moisture.
Reconstitution. Bacteriostatic or sterile water is introduced slowly down the inside wall of the vial. Do not inject the stream directly into the powder and do not shake.
Dissolving. Swirl gently until clear. Peptides are shear-sensitive; vigorous agitation can degrade the compound.
After reconstitution. Refrigerate at 2–8 °C. Reconstituted peptide has a materially shorter working life than the lyophilised form.
Freeze–thaw. Repeated cycles degrade peptides. Aliquot before freezing where a protocol requires multiple uses.

Central rather than vascular action

Unlike compounds that act on blood flow, PT-141 works through melanocortin receptors in the central nervous system. That difference in mechanism is the reason it was developed and the focus of most of the research. 12

MC3R and MC4R selectivity

Changing the C-terminal amide of Melanotan II to a free acid reduces activity at MC1R, the pigmentation receptor, and shifts the profile toward MC3R and MC4R. It is a textbook demonstration of how one terminal change redirects receptor selectivity.

Melanocortin receptor mapping

Used alongside Melanotan II in binding assays to determine which receptor subtype is responsible for which observed effect, since the two differ at a single position.
